EFFECT OF TETRACYCLINE GROUP ANTIBIOTICS ON FIBROBLAST PROLIFERATION AND DNA CONTENTS IN VITRO

Abstract
This study was performed to evaluate the direct effect on minocycline, doxycycline and tetracycline belong to tetracycline group antibiotics, which were well known to enhance gingival reattachment on periodontal tissue. Cellular multiplying activity and cell proliferation were observed on human gingival fibroblasts exposed to above three tetracycline group antibiotics in vitro at a concentration range from 20 to 200§­/ml.
1. At a dose of 20§­/ml, fibroblast numbers of exposed group to minocyclin, doxycycline and tetracycline were similarly increased but a little lower than control group. Minocycline showed maximun proliferation while tertacycline showed the least proliferating at a dose of 50§­/ml. Minocycline and doxycyline enhanced similar proliferation but tetracycline has no effect in fibroblast proliferating activity at a dose of 100§­/ml. Proliferating activity of fibroblast was decreased in all groups at 6 hours after exposure to antibiotics at a dose of 200§­/ml.
2. On measurement of DNA contents by flow cytometry. S-G_2M fractions of cells exposed to minocycline and doxycycline was similar to control group at a dose of 20§­/ml. That of fibroblasts exposed to minocycline and doxycycline was increased, especially minocycline group showed significant increase at a dose of 50§­/ml. All groups exposed to antibiotics showed lower than control group of S-G_2M fraction at dose of 100 and 200§­/ml. There was no significant difference among three groups exposed to antibiotics at a dose of 200§­/ml.
